Amyloid beta (Aβ or Abeta) denotes peptides of 36–43 amino acids that are crucially involved in Alzheimer's disease as the main component of the amyloid plaques found in the brains of Alzheimer patients. The peptides derive from the amyloid precursor protein (APP), which is cleaved by beta secretase and gamma secretase to yield Aβ. Aβ molecules can aggregate to form flexible soluble oligomers which may exist in several forms. It is now believed that certain misfolded oligomers (known as "seeds") can induce other Aβ molecules to also take the misfolded oligomeric form, leading to a chain reaction akin to a prion infection. The oligomers are toxic to nerve cells. The other protein implicated in Alzheimer's disease, tau protein, also forms such prion-like misfolded oligomers, and there is some evidence that misfolded Aβ can induce tau to misfold. β-淀粉樣蛋白來(lái)自β-淀粉樣蛋白原，在腦組織的細(xì)胞外呈絲狀蛋白樣沉積 ，是淀粉樣結(jié)節(jié)性神經(jīng)炎病變的主要蛋白成分，在神經(jīng)纖維中也有沉積。 在老年性癡呆Alzheimer病中，大腦皮質(zhì)中特征性地出現(xiàn)β-淀粉樣蛋白沉積形成的老年斑。 溫馨提示：β淀粉樣肽溶解較困難，建議先用少量DMSO溶解，再緩慢加入1×PBS，邊加邊混勻。注意不要使用混勻器進(jìn)行操作。 |
