細(xì)胞周期是細(xì)胞生命活動(dòng)的基本過程,細(xì)胞在周期時(shí)相的變遷中進(jìn)入增值、分化、衰老和死亡等生理狀態(tài)。細(xì)胞周期分為間期和分裂期:間期又分為三期、即DNA合成前期(G1期)、DNA合成期(S期)與DNA合成后期(G2期);分裂期(M期)又分為前期,中期,后期和末期。
研究發(fā)現(xiàn)在細(xì)胞周期中起非常重要的幾類調(diào)控因子:一類是細(xì)胞生長(zhǎng)因子對(duì)細(xì)胞分裂增殖有調(diào)控作用,如血小板衍生生長(zhǎng)因子PDGF、轉(zhuǎn)化生長(zhǎng)因子TGF等;另一類是細(xì)胞周期調(diào)控因子,包括細(xì)胞周期蛋白cyclin、細(xì)胞周期蛋白依賴激酶CDK、CDK抑制因子CKI和促進(jìn)CDK調(diào)節(jié)因子降解的酶復(fù)合物(如SCF和APC酶復(fù)合物)。
由于細(xì)胞周期的失調(diào)與腫瘤的發(fā)生密切相關(guān),同時(shí)細(xì)胞周期調(diào)控因子也參與體內(nèi)多種細(xì)胞分化,因此目前對(duì)細(xì)胞周期調(diào)控的研究也是生物學(xué)研究的熱點(diǎn)課題,更深更廣地探討分子間相互作用和基因表達(dá)調(diào)控的機(jī)制,為以后腫瘤的治療提供新的思路。
